The FHF complex is inherently autoinhibited through HOOK3 intramolecular interactions. b , KIF1C, which is autoinhibited, binds (through its stalk domain) to the HOOK3 C terminus and relieves the HOOK3 fold-back conformation. This enables dynein–dynactin access to the HOOK3 N terminus and minus end motility. During these runs, KIF1C only partially engages the microtubules, acting as a processivity factor for dynein. c , Dynein–dynactin, at sufficiently high local concentration, binds HOOK3 at its N terminus, exposing the C terminus for KIF1C binding and activation. Dynein weakly engages microtubules and slows down plus end transport.
